| مقدمه: ذرت خوشهای (Sorghum bicolor L.) از مهمترین غلات در سراسر جهان است که بهعنوان غذای انسان و علوفه دام استفاده میشود. تنش شوری نیز از مهمترین تنشهای غیر زیستی میباشد که موجب کاهش رشد و عملکرد گیاهان میشود. بر این اساس این تحقیق بهمنظور بررسی تغییرات بیوشیمیایی در دو رقم متحمل و حساس ذرت خوشهای تحت تأثیر شدت و مدت زمان اعمال تنش شوری و تجزیه و تحلیل الگوی بیان ژنهای منتخب خانواده bZIP و ارتباط آنها با سازوکارهای دفاعی گیاه در ذرت خوشهای اجرا گردید. مواد و روشها: این آزمایش به صورت طرح فاکتوریل اسپلت پلات در قالب بلوکهای کامل تصادفی با سه تکرار در سال 1400 بهصورت آزمایش گلدانی در گلخانه دانشکده علوم کشاورزی دانشگاه گیلان اجرا شد. تنش شوری شامل چهار سطح شاهد (آب)، 75، 125 و 175 میلیمولار کلرید سدیم بود که در پایان مرحله 5-4 برگی اعمال شد. نمونههای برگی در 24، 48، 72 و 96 ساعت پس از اعمال تنش جمعآوری شدند. سپس صفتهایی مانند پایداری غشای سلولی، پروتئینهای محلول، فعالیت آنزیم کاتالاز و فعالیت آنزیم سوپراکسیددیسموتاز ارزیابی شدند. میزان بیان ژنها نیز از طریق روش Real-Time PCR در ارقام متحمل و حساس بررسی شد. یافتهها: نتایج نشان داد که با افزایش شدت تنش شوری فعالیت آنزیم کاتالاز و سوپراکسید دیسموتاز و نشت یونی روند افزایشی داشتند. در این شرایط فعالیت آنزیم کاتالاز و سوپراکسید دیسموتاز به ترتیب 7/2 و 9/4 برابر و نشت یونی 75 درصد افزایش یافت. محتوای پروتئینهای محلول نیز (55 درصد) تحت شرایط تنش شوری به طور معنیداری کاهش یافت. همچنین نتایج بررسی میزان بیان ژنهای SORBI_3002G225100، SORBI_3003G332200، SORBI_3003G368300، SORBI_3010G081800 و SORBI_3004G293500 نشان داد که بین زمانهای مختلف نمونهبرداری پس از اعمال تیمار تنش شوری در ارقام حساس و متحمل تفاوت معنیداری وجود داشت و میزان بیان نسبی ژنهای مورد بررسی در ارقام متحمل و حساس در طول تنش شوری روند افزایشی داشت. نتیجهگیری: بر اساس نتایج بهدست آمده میتوان دریافت که ذرت خوشهای بهعنوان یک گیاه متحمل به شوری از سازوکارهایی همچون افزایش بیان ژنهای مرتبط با تنش و افزایش فعالیت آنزیمهای آنتیاکسیدان برای افزایش تحمل به تنش شوری استفاده میکند. این تحقیق همچنین نشان داد که رقم متحمل فومن بهویژه در شرایط تنش شوری عملکرد بهتری نسبت به رقم حساس از خود نشان داد. بهطوری که فعالیت آنزیمهای آنتیاکسیدانی و بیان ژنهای مرتبط با تحمل شوری در این رقم بیشتر بود. در نتیجه، استفاده از ارقام متحمل به شوری میتواند بهعنوان یک راهکاری مفید در بهبود تحمل به تنشهای محیطی در گیاهان معرفی شود. | ||
