| مقدمه: شوری خاک همچنان یکی از چالشهای اصلی امنیت غذایی جهانی است که بهرهوری محصولات کشاورزی را در گونههای مختلف بهشدت تحت تأثیر قرار میدهد. با وجود مطالعات متعدد پیرامون تحمل به شوری در گیاهان زراعی مختلف، بررسیهای تطبیقی بینگونهای همچنان محدود است. این پژوهش با هدف پر کردن شکاف میان پاسخهای مولکولی و صفات مرفوفیزیولوژیک از طریق شناسایی مکانیسمهای پاسخ به شوری (مشترک و اختصاصی گونه) در برنج (Oryza sativa) و گوجهفرنگی (Solanum lycopersicum) طراحی شد. هدف نهایی این مطالعه، اولویتبندی ژنهای کلیدی و مسیرهای متابولیکی است که میتوانند بهعنوان اهداف قابلاطمینان در برنامههای اصلاح نژاد دقیق مورد استفاده قرار گیرند. مواد و روشها: در این مطالعه از رویکرد متاآنالیز دادههای ترانسکریپتومیک (ماکرواری) استخراجشده از پایگاه داده NCBI GEO استفاده شد. دو مجموعه داده انتخاب گردید: GSE16401 (گوجهفرنگی: رقم متحمل PL36 در مقابل رقم حساس Money Maker) و GSE58603 (برنج: رقم متحمل PL177 در مقابل رقم حساس IR64). تحلیل بیان افتراقی ژنها (DGE) با استفاده از ابزار GEO2R و با اعمال آستانه سختگیرانه |log2FC| ≥ 1 و FDR ≤ 0.05 انجام گرفت. ژنهای بیانیافته افتراقی (DEGs) مشترک و اختصاصی از طریق ابزار Venny 2.1.0 شناسایی شدند. در ادامه، برای روشن شدن نقشهای بیولوژیکی و چارچوبهای متابولیکی درگیر در تحمل به شوری، تحلیل غنیسازی عملکردی (GO و KEGG) با استفاده از پایگاه WebGestalt 2024 صورت گرفت. یافتهها: نتایج نشاندهنده یک الگوی ترانسکریپتومیک متمایز بین ژنوتیپهای متحمل و حساس در هر دو گونه بود. در ژنوتیپهای متحمل، افزایش بیان هماهنگ ژنهای مرتبط با بازسازی دیواره سلولی، تنظیم اسمزی و متابولیسم ثانویه مشاهده شد. بهطور خاص، انتقالدهندههای یونی کلیدی نظیر خانوادههای HKT و NHX، در کنار ژنهای دفاع آنتیاکسیدان، بیان افتراقی معنیداری را نشان دادند که بر نقش آنها در حفظ هموستاز یونی و کاهش خسارات اکسیداتیو تأکید دارد. علاوه بر این، چندین خانواده فاکتور رونویسی شامل DREB، NAC و WRKY بهعنوان تنظیمکنندههای مرکزی پاسخ به شوری شناسایی شدند. در مقابل، ژنوتیپهای حساس کاهش بیان شدیدی را در ژنهای مرتبط با فتوسنتز، متابولیسم کربن و پیامرسانی هورمونی نشان دادند که میتواند نشاندهنده فروپاشی هماهنگی فیزیولوژیک تحت تنش شوری شدید باشد. نتیجهگیری: یافتههای این پژوهش، چشمانداز جامعی از مکانیسم مولکولی حاکم بر تحمل به شوری در دو محصول با تبار تکاملی متفاوت ارائه میدهد. شناسایی مسیرهای مشترک (بهویژه آنهایی که با ظرفیت آنتیاکسیدانی و تنظیم اسمزی مرتبط هستند) نشاندهنده وجود یک «جعبهابزار» بنیادی برای سازگاری با شوری در گیاهان است. این نتایج کاندیدهای مولکولی ارزشمندی را جهت ارتقای تحمل به شوری از طریق انتخاب ژنومیک یا ویرایش ژنی فراهم کرده و نقشه راهی برای توسعه ارقام مقاوم در برابر شرایط محیطی نامساعد در غلات و صیفیجات ارائه میدهند. | ||
